The answer to your question is yes as the turnbuckles are stronger by far than the wire. However it strikes me that stretching the wire over that length seems a bit ambitious as it is longer than usually the case so it would have to be tensioned more than normally done. Depends a bit on what the wire is meant to do. It may not hold back a raging bull but could hold up a canvas banner. It could be tightened enough to break the wire.

No, you turn the middle section (after unscrewing either end first of all) and this will tighten each bit without twisting the wires. Should not break if a decent wire.
