Not quite sure what you mean by steering knuckle but providing you have a fixed point for the centre piston in relation to the disc then yes. This is a pretty powerful bit of kit.

It would. But depending on the vehicle, the disc should tap off with a soft mallet.

How much clearance will I need at the side of my wheel hub in order to attach and pump this thing?

If you mean how much space at the side of the car, I would say at least a metre and more space in order really put some strength into the lever.
