If i pack this say half way full, could it be used as a carry on bag for a airplane trip?

Typical allowed size is 9" x 14" x 22". This bag is 12" x 12" x 37" so you'd be over 9" for the one dimension unless you only packed it 9" full or didn't fill it to the sides. I think it would really depend on the carrier since it's a flexible canvas but the bag and straps are bulky. I think many airlines would allow it as long as you could squish it down to the allowed size, but you won't really know until you try.

Is it possible to use a padlock to protect the main compartiment?

There's only one compartment it's a military C-bag and yes you can use a padlock.

How much weight would this bag be able to hold without ripping after several uses?

It easily holds around 50-55 lbs, but that is not a concern for failure. It is cloth and it is susceptible to cuts by a sharp object (which is what happened to mine).

id like to end up with a 54"x32" GI style duffel bag with straps to carry on back

This was a gift for my granddaughter. She was traveling to Cambodia for school. It did remind me a lot of the bag my husband in the army years ago. Plus it was waterproof.
