Will it stick to a boot that has a plastic boot liner?

Because of the design of the product it doesn't move when in place in your vehicle. And of course when you have items in there it adds to the weight as well. I use it for carrying all my work needs. It keeps everything in one place but with the removable separators it keeps everything neat and easy to access

Note that there are three compartments - can this be changed (remove the small divider) so that are just two equal sized ones?

Yes the small divider can be left out. It is fitted using a velcro strip.

Can you remove all the compartments so it is just a single box ?

Only one of the compartments to make it the same size .
